Cells 'flock' to heal wounds: Researchers analyze physics of epithelial cell cooperation

Jan. 24, 2013 ? Like flocks of birds, cells coordinate their motions as they race to cover and ultimately heal wounds to the skin. How that happens is a little less of a mystery today.

Researchers once thought only the cells at the edge of a growing patch of wounded skin were actively moving while dividing cells passively filled in the middle. But that's only part of the picture. Rice University physicist Herbert Levine and his colleagues have discovered that the process works much more efficiently if highly activated cells in every part of the patch exert force as they pull their neighbors along.

There's a need to understand how cells cooperate to protect the site of a wound in the hours and days after injury, said Levine, who has introduced the first iteration of a computer model to analyze the two-dimensional physics of epithelial sheets. He hopes it will give new insight into a process with long-term implications not only for healing but also for understanding cancer, a prime motivator in his research since joining Rice under a grant from the Cancer Research and Prevention Institute of Texas.

A paper on the research by Levine, based at Rice University's BioScience Research Collaborative, and colleagues at the University of California at San Diego and in Germany and France appears January 23 in the Proceedings of the National Academy of Science.

Levine and his colleagues create computer models of processes seen by experimentalists to flesh out the rules that govern biological systems. "Here, we're combining experimental observations from single cells with general notions from the physics literature to create an integrated way of thinking about this multicellular system," he said.

The new models were prompted by a recent Harvard study showing "that even in the middle of a sheet, cells were dynamically creating heads and tails and were actively moving rather than being passively carried along," Levine said. "This data convinced us that we needed a different way to start to think about the problem."

The body marshals an astounding array of forces to heal wounds, Levine said. Many have to do with cell biology, the internal and external signals that tell a cell when to move, when to stop, when to split and when to die. His team's intent was to focus first on the cell's physical interactions with its neighbors and study what happened if all those complicating factors are eliminated from the simulations.

"We try to unravel what is physics and what is biology," Levine said. "We want to know which parts of the phenomenon don't require sophisticated signaling networks."

In the physics approach to cell motility, he said, "the first thing to do is see how far we can get if we assume that all the cells are following the same rules. Then the only thing that's creating the dynamics of the system is that they're interacting with each other. This is the type of problem that physicists have studied before, usually in nonbiological contexts."

In the Harvard experiment, he said, "They had taken a millimeter-sized tissue that was spreading and showed it wasn't just cells on the end that were pulling on the tissue while the others were spectators." But that work didn't explain how cells in the center of the tissue knew the direction of the edge.

Levine's team looked to the skies for inspiration. "Birds look around and decide which way all their neighbors are flying," Levine said. "The idea that they would move as independent birds but also coordinate is where the idea of flocking came from. This way of thinking hadn't been applied to epithelial tissue motility in wound healing."

What cells "see" are their sticky neighbors, which pull and tug them as they move on lamellipodia, thin sheets that serve as "feet" powered by actin filaments that act something like the treads on a tank. The overlapping lamellipodia of adjacent cells influence each other. "The cells have to figure out which way to go based on competing tendencies: their own tendency to push on the ground and propel themselves forward, and the tendency of their neighbors to try to pull them in various directions," Levine said. "Our basic notion is that as time goes on, these tendencies become correlated as the cell 'tries' to accommodate its conflicting inputs."

The Harvard experimentalists saw that loosely packed cells in the middle of a growing colony tend to swirl in a disorganized manner, and the simulations confirmed this. These swirls are analogous to what is seen in other examples of flocking. But when a wound is introduced, the swirls disappear and cells begin to match direction and velocity and pull toward a common goal. The ones on the edge immediately know which way to go, and everyone else learns from their example. Surprisingly, Levine said, "stickier" cells tend to push forward unevenly, with finger-like protrusions at the leading edge, much like what experimentalists often see.

The simulation model has a long way to go, Levine said. "It's rough around the edges. Biologists who read this will immediately say, 'You've left out all sorts of interesting things we know are happening.'

"Yes, there will be experiments for which this approach will not be sufficient," he said. "It will teach us that in those cases, biology has to exert a more specific role in creating the structures and the motion."

Levine hopes to match the models to current work by experimentalists on motility in cells related to the metastatic spread of breast cancer. "We're a long way from saying anything about this problem," he said. "But that's my overall agenda -- to push my research to where it can make contact with the cancer community."

Co-authors of the new paper are postdoctoral researcher Markus Basan and senior scientist Wouter-Jan Rappel of the Center for Theoretical Biological Physics, University of California at San Diego; Jens Elgeti, a member of the scientific staff at the Institute of Complex Systems, J?lich, Germany; and Edouard Hannezo, a graduate student at the Curie Institute, Paris. Levine is the Karl F. Hasselmann Professor in Bioengineering at Rice and co-director of the Rice-based Center for Theoretical Biological Physics.

Plane with 3 aboard missing near South Pole

An aircraft carrying three men went missing in Antarctica on Monday and the plane?s emergency locator beacon was activated, according to the National Science Foundation. ?

The status of those aboard remains unknown, NSF spokesman Peter West said Wednesday.?The Toronto Star reported that the three men are Canadian.

West said those aboard were likely a pilot, co-pilot and a flight engineer carrying or picking up cargo. They had been contracted to support a branch of the?Italian Antarctic Program that focuses on new technology and energy?? specifically nuclear fusion and fission.

The aircraft, a de Havilland Twin Otter, was returning from the South Pole to Terra Nova Bay, where the Italian Antarctic Program is based, when contact was lost as the plane flew over a remote area of the Transantarctic Mountains.


The plane was contracted out by Kenn Borek Air Ltd., a Canadian company based in Calgary that charters aircraft to the U.S. Antarctic program.

Rescue crews, based at the New Zealand Rescue Coordination Center, know generally where the beacon is coming from, but cloudy and windy conditions have prevented rescue planes from attempting a landing near the downed plane.

?There are not as many weather stations, so it?s difficult to find out what the weather is,? West said. ?There was low cloud, limited visibility in the air in the area where they were looking for the aircraft -- some blowing snow and issues with cloud.?

West said he didn?t know whether the flight crew carried cold weather survival gear. At the U.S. station, protocol demands that anyone leaving the base must have protective gear ? typically a parka, wind pants, insulated boots, a tent, food and a stove to melt snow into water.

He said that he doesn't recall a similar crash in his 14 years as a spokesman for the Antarctic program.

Antarctica, the size of U.S. and Mexico combined, is vast, white and isolated. There are about 50 research stations, some of them year-round, others open during research season, which runs roughly between October and early February ? summer in the Southern Hemisphere. During those months, the largest is McMurdo Station, the U.S. Antarctic station on Ross Island, with about 1,100 people.

?It?s a harsh continent,? West said. ?People take extra care if they can.?

Boehner Pressures Senate to Pass Budget or Have Pay Withheld

WASHINGTON - Despite President Obama's call for Congress to prioritize issues like immigration reform and address climate change during his inaugural address Monday, House Speaker John Boehner said that the House of Representatives will instead continue its focus on fiscal responsibility, including a plan to balance the budget over the next decade.

"There are a lot of priorities for the Congress [and] a lot of priorities for the president, but right now the biggest issue is the debt that's crushing the future for our kids and our grandkids," Boehner, R-Ohio, said. "Taxpayers understand that you can't keep spending money that you don't have, so we're going to continue to focus, especially here over this next 90-, 120-day period, on bringing some fiscal responsibility to Washington."

Boehner, who conducted his first news conference since Dec. 21 this evening, said the House will move forward on its No Budget No Pay act, which directs both chambers of Congress to adopt a budget resolution for FY 2014 by April 15, 2013. If either body fails to pass a budget, members of that body would have their paychecks put into an escrow account starting on April 16 until that body adopts a budget. But, because of the 27 th Amendment, any pay that is withheld would eventually be released at the end of the current Congress, even if a budget doesn't ever pass.

"Over the last four years, House Republicans have offered plans, our budget plans. We've done our budgets but it's been nearly four years since the Senate has done a budget," he said. "Most Americans believe you don't do your job you shouldn't get paid. That's the basis for No Budget, No Pay. It's time for the Senate to act."

The bill, H.R. 325, also temporarily suspends the statutory debt limit until May 18, granting the Treasury Department the additional borrowing authority to meet obligations that require payment over the next three months.

Facing trillion dollar annual budget deficits, Boehner told the House Republican Conference this afternoon that the GOP will advance a budget that becomes balanced over the next 10 years.

"It's time for us to come to a plan that will in fact balance the budget over the next 10 years," he added. "It's our commitment to the American people and we hope the Senate will do their budget as they should have done over the last four years."

Still, the country's growing $16 trillion federal deficit would not balance by then, and Boehner stressed that Republicans are not open to new tax revenue as part of a comprehensive plan to reduce the deficit.

"We are not going to raise taxes on the American people," Boehner insisted. "Nobody on our side is interested in raising taxes on the American people. It's time to deal with the serious problem that we have, which is spending."

That is sure to be a point of contention among Democrats. House Minority Whip Steny Hoyer rejected the GOP's doggedness that new revenue would not be part of a package to reduce the deficit.

"We absolutely reject that. The president absolutely rejects it, I reject it, period," Hoyer, D-Md., said during a pen and pad briefing with reporters Tuesday. "Going forward, revenues are going to still need to be part of the solution, as are cuts and restraints and reduction of growth. All are going to have to be part of it."

The House is expected to vote Wednesday on a three-month increase to the debt limit, a move intended to sync up the next slate of fiscal deadlines facing Congress and provide lawmakers with more time to work out a so-called "Big Deal" on deficit reduction.

As he left the news conference, Boehner ignored a question about whether he believes the measure will pass. Still, the president has promised not to veto it and many congressional Democrats appear open to supporting it.

"This bill is a game bill. It's not a substantive response to a serious problem," Hoyer said. "Having said that, I've made it very clear that defaulting is not an option. All the Republicans leaders believe it's not an option. And so I think, in that context, I'll have to look at it."

4. ?Ask if you can stay in touch.? Get permission and then give your word that you will do it.

5. ?Say thank you. Send them a handwritten note thanking them for taking time to reconnect.

6.? Just do it!? Stay in touch using a simple contact management system.

Remember that each loyal customer or contact in your database can represent a transaction down the line. ?If you?re not keeping in touch with them and building relationships after the sale, you?re missing out on opportunities to move your business forward.

Bloggers Put Chemical Reactions through the Replication Mill

Scrounging chemicals and equipment in their spare time, a team of chemistry bloggers is trying to replicate published protocols for making molecules. The researchers want to check how easy it is to repeat the recipes that scientists report in papers ? and are inviting fellow chemists to join them.

?We?re just a bunch of people who want to make the reactions work,? explains blogger See Arr Oh, who is based in the United States and prefers not to reveal his real name. The other team members include chemistry graduate student Matt Katcher from Princeton, New Jersey, and two bloggers called Organometallica and BRSM, who together have launched Blog Syn, in which they report their progress online.?

Among the frustrations he and others have experienced with the chemical literature, says See Arr Oh, are claims that reactions yield products in greater amounts than seems reasonable, and scanty detail about specific conditions in which to run reactions. In some cases, reactions are reported which seem too good to be true - such as a 2009 paper which was corrected within 24 hours by web-savvy chemists live-blogging the experiment; an episode which partially inspired Blog Syn. ?I never forgot that reaction,? See Arr Oh adds.?

?Ask any synthetic chemist what they spend most of their time on, and they will admit that it is getting literature reactions to work,? says chemist Peter Scott of the University of Warwick in Coventry, UK. ?That is the elephant in the room of synthetic chemistry.?

Blog Syn is one of several places where chemists can check synthesis know-how. The journal Organic Syntheses, for example, only publishes synthetic procedures that have been checked and verified by a member of its editorial board. Scott is involved in a separate project to compile a database of about 5,000 chemical procedures, called SyntheticPages, which is part of ChemSpider, an online database of chemical structures.? And the Journal of Visualized Experiments (JoVE) ? a peer-reviewed online journal that publishes videos of experiments ? will launch a chemistry channel on 4 February. That could help with one difficulty faced by some chemists: published chemistry reactions are written in a standard format that is supposed to make things easy to follow, yet unwritten tweaks are often needed to make experiments succeed in practice.

See Arr Oh says that Blog Syn is different to JoVE and SyntheticPages because those initiatives involve chemists refining and verifying their own chemical reactions. By contrast, Blog Syn intends to use crowd sourcing, meaning that outsiders ? chemists that the bloggers already know, to start with ? will check work that has already been published by others.

Test case
So far, the bloggers have investigated one reaction, which they picked because it would be especially useful if it were easy to replicate; it uses an iron and sulphur catalyst to make ring-shaped molecules containing oxygen and nitrogen. Following the published method, the team couldn?t get as much of the final product as the original authors did. ?It does indeed work, but we?re not positive yet what factors the authors have that we don?t,? says See Arr Oh. One problem, the team's blogpost says, was that the written paper didn't make it clear how stringently they needed to exclude air from the reaction.

The chemists? online efforts reflect a growing focus across the sciences on the importance of being able to reproduce published procedures. One project, the Reproducibility Initiative, was set up last year to enable researchers to have their study ?validated? by outside checkers. Another, the Reproducibility Project, is dedicated to estimating the reproducibility of psychology experiments. The director of that project, social psychologist Brian Nosek of the University of Virginia in Charlottesville, thinks that Blog Syn will work only if chemists can be rewarded for participating.

See Arr Oh accepts that funding Blog Syn will be the challenge to developing it on a larger scale. He is exploring ways to make the blog easier to manage, including crowd-sourced funding methods such as the website Kickstarter. Until then, he will continue to check reactions in his spare time. ?At the moment these reactions are done at night under cover of darkness,? he says.

Obama Says Country, As 'One People,' Must Tackle Immigration ...

President Barack Obama used his inauguration speech Monday to ask Americans to come together to tackle the country?s toughest challenges, including immigration reforms, protecting voter rights, and gun law changes.

"My fellow Americans, we are made for this moment, and we will seize it ? so long as we seize it together," Obama told a crowd of an estimated 800,000 who gathered on the National Mall to see the 57th Inauguration amid overcast skies and chilly winds.

Obama's speech focused on the power of Americans to solve the nation's hardships together. The country, he said, had come through a Civil War that helped end slavery; worked together to build railways and highways; built schools; regulated the free market; and cared for the vulnerable.

"But we have always understood that when times change, so must we; that fidelity to our founding principles requires new responses to new challenges; that preserving our individual freedoms ultimately requires collective action," Obama said. "Now, more than ever, we must do these things together, as one nation and one people."

In 18 minutes, Obama stuck to broad concepts of citizenship and stayed away from details. Near the end, however, he used one sweeping stanza to touch on a range of issues that are taking shape as the most pressing issues of his second term?including immigration, gun control, and voting rights. In a broad sense, he said, he said it was this generation's task to carry on the idea of freedom from the pioneers.

"For our journey is not complete until our wives, our mothers and daughters can earn a living equal to their efforts," Obama said. "Our journey is not complete until our gay brothers and sisters are treated like anyone else under the law ? for if we are truly created equal, then surely the love we commit to one another must be equal as well.

"Our journey is not complete until no citizen is forced to wait for hours to exercise the right to vote," Obama said. "Our journey is not complete until we find a better way to welcome the striving, hopeful immigrants who still see America as a land of opportunity, until bright young students and engineers are enlisted in our workforce rather than expelled from our country," Obama continued.

"Our journey is not complete until all our children, from the streets of Detroit to the hills of Appalachia, to the quiet lanes of Newtown, know that they are cared for and cherished and always safe from harm," Obama said.

Obama also called on Americans to respond to the threat of climate change, saying "that the failure to do so would betray our children and future generations." Americans "cannot afford to delay," he said, and should move forward with "common effort and common purpose."

In recent weeks, Obama has offered proposals on gun law reforms, and has made overtures that immigration could become a centerpiece of his second term.
